NOVN/AMGN/IONS: LP(a) after the pelacarsen failure

1 min read

Pelacarsen’s HORIZON failure materially raises the burden of proof for the Lp(a) class, but we think the investment question for Amgen now shifts from the headline failure to whether the eventual dataset supports a class-wide read-through or leaves room for olpasiran differentiation. Our latest work frames the key clinical and trial-design debates that will determine how much of the HORIZON outcome should be reflected in Amgen’s risk–reward ahead of OCEAN(a), and where current positioning may be too aggressive in either direction.

Our latest report addresses the following key questions:

  • Does HORIZON challenge Lp(a) as a therapeutic target, or is there still a credible case for differentiation within the class?

  • What findings from HORIZON would most materially change the probability of success for OCEAN(a)?

  • Could differences in trial design materially affect the read-through from pelacarsen to olpasiran?

  • Has the market appropriately recalibrated Amgen’s Lp(a) risk–reward following the HORIZON headline failure?
